Pettifer, who sat out the 2019 season following a knee injury, played an assistant role with the club's A grade side this year to outgoing coach Chloe Kavanagh.
The new coach said she was excited to be taking on the role.
“There is a lot of excitement and hype at the club as we enter our 100th year, so I thought, 'what better time to take on the role?'" she said.
“I've been very lucky this year to have been around the group, getting to know the players and seeing them develop so I have a good understanding of our team, so it was a bit of a no-brainer in the end to take on the role.”
Pettifer's Girgarre side will take on a new look for the 2020 season, with the club already active in its recruiting for the new year.
“We've recruited Holly Butler from Kyabram,” she said.
“Holly is originally a Girgarre girl, her family live in Girgarre and her brother plays football for the club, so it was a great moment for us to have her decide she wanted to come back and be a part of the team.
“Bec Squires is joining us from Lancaster, which will be a great addition to our defensive end, and with myself playing and a full season from Sheridan Deledio added to the quality we already have, we will be able to really strengthen the side and add a bit of depth.
“There is a possibility of being able to field a C reserve side for the club as well, which with our already strong junior ranks is a huge effort for our club."
