Star legspinner Adam Zampa has been summoned straight back into the XI for Friday's clash, while seamer Michael Neser has been handed his first cap of the tour.
Zampa, who captured 4-48 in the second game at Bloemfontein, was sorely missed in Potchefstroom as the Proteas' spinners masterminded a landslide 111-run victory to trim Australia's series lead to 2-1.
Zampa comes in for fellow wrist spinner Tanveer Sangha, who was expensive in his one-day international (ODI) debut, while Sean Abbott, who has played all six Twenty20 and ODI fixtures since arriving, has been given a well-earned rest.
Aiden Markram, coming off a match-winning century, will captain South Africa in the absence of regular skipper Temba Bavuma, who is nursing a right adductor strain.
Without Bavuma, the Proteas' leading scorer for the series with 217 runs at 108.50, much will depend on Markram at the top, along with in-form Quinton de Kock and Rassie van der Dussen, who is back after being dropped for the third ODI.
Pace spearheads Kagiso Rabada and Lungi Ngidi return to the team, replacing Sisanda Magala, who has a knee niggle, and left-arm spinner Tabraiz Shamsi, who is being rested.
