Omar Elomar, 18, was shot in the street during an altercation in Miller on February 15, 2020.
Homicide Squad detectives arrested four men, aged 20, 22, 23 and 25, at homes in Bonnyrigg, Macquarie Fields, Gregory Hills and Orange just before dawn on Thursday.
They are expected to be charged over the teenager's death.
In February 2020, Homicide Squad Commander Danny Doherty said the killing had stemmed from a "trivial" interaction.
"It's nothing to do with any other family members, except for the prior incident the day before involving a young teenager who was involved in an altercation, and that's where it led from," Detective Superintendent Doherty said.
Police will speak to the media later on Thursday.
An 18-year-old man previously charged in connection with the murder was acquitted.
