Police said a B-double truck and a medium rigid truck crashed head-on in wet conditions on the Midland Hwy just east of the Turnbull Rd intersection at about 11am.
The driver of one of the trucks was trapped in their vehicle for about an hour.
The 35-year-old Ardmona man was airlifted to a Melbourne hospital by air ambulance with serious, non-life-threatening injuries, police said.
The air ambulance took off from near the crash site shortly before 12.50pm.

According to police, the driver of the other vehicle was not seriously injured; however, they were taken to Goulburn Valley Health by road ambulance.
A section of the Midland Hwy between Turnbull Rd and Excelsior Ave was closed to traffic, with detours in place.
Motorists were being diverted via Excelsior Ave, on to Macisaac Rd, before rejoining at Ardmona Rd outside Mooroopna.
As of 12.50pm, there was no estimate of when the road would be cleared for traffic again.
